Improving effect of domestic walnut (Juglans regia) extract on high glucose- and oleic acid-induced non-alcoholic fatty liver disease in HepG2 cells

This study was investigated to evaluate the in vitro anti-diabetic activity and lipid accumulation inhibitory effect of domestic walnut (Junglans regia, Gimcheon 1ho cultivar) high glucose- oleic acid-induced hepatic HepG2 cells. Eighty percent ethanolic extract from (GC) showed higher effects against α-amylase α-glucosidase activities when compared other extracts. GC significantly inhibited formation advanced glycation end products (AGEs). The increased cell viability H2O2- glucose-induced death determined by 3-(4,5-dimethyl-thiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay. reduced reactive oxygen species (ROS) cells 2’,7’-dichlorofluorescein diacetate (DCF-DA) AMP-activated protein kinase (AMPK) phosphorylation decreased expression metabolism related proteins such as sterol regulatory element-binding 1 (SREBP-1), fatty acid synthase (FAS), 2 (SREBP-2) 3-hydroxy-3-methyl-glutaryl-coenzyme A (HMGCR). results suggest is a potentially functional food material which can improve nonalcoholic liver disease inhibiting accumulation.
